Reliable Genomic DNA from Multiple Blood Sample Types

The InnoSpinM Blood DNA Extraction Kit is designed for the isolation of total genomic DNA from whole blood, plasma, serum, buffy coat, and lymphocytes using a standard microcentrifuge. The silica spin-column format selectively binds DNA under optimised buffer conditions, while sequential wash steps remove contaminants and PCR inhibitors that would otherwise compromise downstream assay performance.

The protocol requires no organic solvents and is designed for routine laboratory workflows. All buffers except Wash Buffer I and Wash Buffer II are supplied ready to use, and kit components remain stable for up to 12 months when stored at 15 to 25°C.

THE PROCESS

Simple Steps Pure Results

HOW IT WORKS

Step 1 – Lysis

Add 20 µL Proteinase K and 200 µL sample to a 1.5 mL microcentrifuge tube. Add 200 µL Lysis Buffer and vortex. Incubate at 56°C for 20 minutes at 1400 RPM.

Step 2 – Binding

Add 200 µL absolute ethanol and vortex. Transfer the lysate to the silica spin column seated in a 2 mL collection tube. Centrifuge at 10,000 RPM for 1 minute.

Step 3 – Wash

Add 500 µL Wash Buffer I. Centrifuge at 10,000 RPM for 1 minute. Add 500 µL Wash Buffer II. Centrifuge at 14,000 RPM for 1 minute. Dry spin at 14,000 RPM for 1 minute to remove residual wash buffer.

HOW IT WORKS

Step 4 – Elution

Transfer the column to a clean 1.5 mL microcentrifuge tube. Add 100 µL elution buffer prewarmed to 56°C directly onto the silica membrane. Incubate for 2 to 3 minutes and centrifuge at 10,000 RPM for 1 minute. Collect purified DNA.

Do any buffers need preparation before use?

Yes. Wash Buffer I and Wash Buffer II are supplied as concentrates and require a one-time addition of 96–100% ethanol before first use. Proteinase K is supplied as a lyophilised powder and must be reconstituted before use. All remaining buffers are supplied ready to use for added convenience.

Does the kit require cold-chain storage?

No. The kit components are stable at room temperature (15–25°C) for up to 12 months from the date of manufacture when stored as recommended. Only the reconstituted Proteinase K requires storage at −20°C to maintain its performance throughout the product's recommended usage period.

How should extracted DNA be stored?

Purified DNA can be used immediately for downstream molecular testing or stored for future use. For long-term preservation, the extracted DNA should be stored at −80°C to help maintain its quality and integrity for subsequent molecular biology applications and repeat analysis when required.
